Wow -- that's a very open-ended question. If I were in your shoes, I'd skip SQL Server 2000 and go straight to SQL Server 2005. You can expect to see major improvements in scalability (the number of users and transactions a single server can support at one time), availability (the amount of time between server outages, either planned or unplanned) and maintainability (the ease with which you can manage the server).
